DARPA aims to write and delete epigenetic base modifications in DNA that could be used for \u201ccognitive enhancement, protection against biological threats, and regenerative medicine<\/em>\u201d.<\/p>\n

According to their announcement, the \u201cDNA-Protein Epigenetic Chemistry Engineerin<\/em>g\u201d (D-PECHE) program of the US Defense Advanced Research Projects Agency (DARPA) aims to \u201cdevelop novel methods for modifying DNA bases in living cells and to validate the presence of new, in vitro catalysed base modifications<\/em>\u201d.<\/p>\n

Instead of permanently altering the DNA, epigenetic modification has the potential to be reversible \u201cwithout changing the underlying genetic sequence<\/em>\u201d.<\/p>\n

Therefore, \u201cintroducing and removing base modifications not found in nature requires new technologies capable of catalysing new chemical modifications of DNA in vitro<\/em>\u201d.<\/p>\n

The Pentagon has long been working on the attempt to create genetically modified super soldiers.<\/p>\n

For example, a Pentagon-funded RAND report published in 2021 outlines the technological possibilities of this controversial transhumanist research, which includes the potential \u201cinsertion of reptilian genes that confer the ability to see in the infrared range<\/em>\u201d, as well as the \u201cenhancement of human powers, increase in intelligence or better adaptation to extreme environments<\/em>\u201d.<\/p>\n

If successful, these \u201cpeople\u201d would have the potential to never tire and to think more intelligently, move faster, jump higher, see farther, hear better, hit harder, live longer, adapt better and calculate faster than any other human being on this planet.<\/p>\n

Now DARPA is turning to epigenetics<\/a> to potentially make processes like some of the above reversible.<\/p>\n

\u201cEpigenetic modifications such as DNA methylation, post-translational modifications of histones and chromatin remodelling can regulate gene expression and DNA-protein interactions without altering the underlying genetic sequence.<\/em>\u201d \u2013 DARPA, D-PECHE program, May 2026<\/p>\n

\u201cDARPA is requesting innovative proposals to demonstrate the feasibility of novel mechanisms for reading and writing epigenetic base modifications in DNA.<\/em>\u201d \u2013 DARPA, D-PECHE program, May 2026<\/p>\n

The main contact person for DARPA’s D-PECHE program is Dr. Aric Lu, who received his PhD in Bioengineering from Harvard University in 2024. Dr. Lu is listed as an Innovation Fellow \u2013 and not as an official program manager \u2013 both on the DARPA website and in his LinkedIn profile.<\/p>\n

According to his DARPA biography, at Harvard University \u201cDr. Lu used genetic engineering techniques to create patterns of cell types and gene expression in bio-printed tissues<\/em>\u201d.<\/p>\n

In a 2023 interview<\/a> with the Humans of Wyss<\/em> series of the Wyss Institute for Biologically Inspired Engineering at Harvard University, Dr. Lu explained what kind of real-world problems he wanted to solve:<\/p>\n

\u201cIf one day we could use genetic engineering and 3D bio-printing to create tissue to repair or replace an organ, it would be very valuable for people with a wide range of diseases, including those on transplant waiting lists<\/em>,\u201d he said, adding: \u201cHowever, to get to the point: We need printed tissue that behaves like human tissue<\/em>.\u201d<\/p>\n

\u201cParticipants in the D-PECHE program need to utilize expertise in nucleic acid chemistry, protein engineering, and synthetic biology to overcome the challenges of enabling and disabling epigenetic modifications. Successfully doing so will reduce the risk associated with a broader concept for epigenetic editing platforms and their potential applications.<\/em>\u201d \u2013 DARPA, D-PECHE program, May 2026<\/p>\n

The D-PECHE study comprises two main tasks:<\/p>\n

    \n
  1. Demonstrate the ability to develop mechanisms for DNA modification to inscribe and\/or delete DNA base modifications. Identify the target modification, catalyst candidates, and the substrates and cofactors required for base modification. Identify any changes to protein sequences or domains required for functionality in living cells.<\/li>\n
  2. Detection of DNA modifications in vitro. Demonstration of the biocompatibility of targeted modifications in a host cell line. Development of assays for the detection of base modifications. Establishment of approaches for the detection of novel modifications in cells.<\/li>\n<\/ol>\n
